dc.description.abstractThe study aims to improve the productivity of cellulase enzyme obtained from bacterial isolates and evaluate enzyme immobilization potential. In the present research work soil samples were collected from three different region of district Jaunpur, Uttar Pradesh i.e. Site-1 Purvanchal University Campus, Site-2 Agricultural field of village Devakali and Site-3 Naveen Sabji Mandi, Chaukiyan. Soil samples were collected from the surface by using a sterile spatula and kept in sterile poly bags and transported to the laboratory. Total 23 bacterial strains were isolated among which three bacterial strains were screened for cellulase production. Morphological test, biochemical test and 16SrDNA gene sequencing were identified bacterial strains as Bacillus paramycoides PS05, Bacillus velezensis PS09 and Bacillus velezensis PS14. Optimization of pH, temperature and carbon sources in culture medium were carried out for maximum cellulase production. The optimum conditions found for maximum cellulase production were 40and#8451; at pH 7 with glucose and Carboxymethylcellulose as carbon source. Among bacterial strains, Bacillus velezensis PS09 is the best cellulase producer followed by Bacillus paramycoides PS05 and Bacillus velezensis PS14. Crude cellulase enzyme is immobilized in calcium alginate bead by entrapment method. The enzyme immobilization efficiency was estimated by the measuring of calcium alginate beads sizes and concentration of enzyme used. The highest enzyme efficiency was shown with 10% concentration and 3.45mm bead size of calcium alginate. newline
