newlineBackground: Helicobacter pylori (H pylori) is a fastidious, microaerophilic bacterium
newlinepresent in the human gastric mucosa of 70-90% of the population in developing
newlinecountries. It plays a significant role in the pathogenesis of chronic gastritis and peptic
newlineulcer disease. Inspite of available diagnostic methods to detect H pylori there is no
newlinesingle technique that can meet the criteria in identification of the bacterium. The cagA,
newlinewhich is not present in every H pylori strain is a marker for pathogenicity island and is
newlineassociated with more severe clinical outcomes. However there are very few studies
newlinelooking directly at the presence of cagA gene in gastric antral biopsies of patients.
newlineTherefore the present study was done to evaluate PCR for detection of H pylori cagA
newlinegene in gastric biopsies as well as their corresponding H pylori isolates, to evaluate
newlinediagnostic tests for identification of H pylori and to assess their antimicrobial
newlinesusceptibility pattern.
newlineObjectives: 1. To study the relative proportion of cagA virulence marker in
newlineHelicobacter pylori isolates and gastric biopsy samples by polymerase chain reaction.
newline2. To evaluate the sensitivity of culture technique for detection of Helicobacter pylori
newlinefrom gastric biopsy samples.
newline3. To generate baseline data on antibiotic sensitivity pattern of H pylori.
newlineMethodology: Biopsies of gastric tissue from 200 patients with disorders of the upper
newlinegastrointestinal tract were studied for H pylori detection by various methods like rapid
newlineurease test, culture, Gram s stain, Giemsa stain, Haematoxylin and Eosin stain,
newlineantibiotic susceptibility testing of the isolated strains, PCR for 16S rRNA and cagA
newlinegene.

newlineResults: Out of 200 patients H pylori was detected by rapid urease test in 26.5%,
newlineGram s staining in 18.5%, Giemsa staining in 29.5%, HandE staining in 14.5%, culture
newlinein 2.5%, PCR for 16SrRNA in 48.5% and PCR for cagA in 31%. H
