Studies on Lipase Producing Probiotic Bacteria as In Vitro Hypocholesterolemic Agent

Abstract

The objectives of this study were: 1) To isolate Lactic Acid Bacteria (LAB) from the newlinedairy samples. 2) To screen lipase producing LAB. 3) To study the in vitro probiotic newlinepotential of LAB such as antimicrobial activity, acid tolerance, bile salt tolerance, newlineantibiotic susceptibility, etc. 4) Determination of Auto-aggregation; cell surface newlineHydrophobicity. 5) To study lysozyme, Pepsin and Pancreatin Resistance. 6) To study newlinethe cholesterol-reducing property of LAB isolates in MRS broth and Simulated intestinal newlinejuice broth. 7) Bile salt hydrolase (BSH) activity by plate assay. 8) Molecular newlineidentification of LAB isolate by 16S r RNA analysis. newlineLactobacillus strains are a major part of the microflora of the gut and of fermented newlinedairy products, and are found in a variety of environments. In this study, the Lactobacillus newlinespecies were isolated from dairy samples collected from traditional milk practicener from newlinegeographic area of North Nanded district. Around 24 isolates from 32 samples were newlineisolated. Among twenty four isolates seven isolates were selected on the basis of lipase newlineactivity. Lipase activity was analysed using tributyrin agar. Lipase producing isolates newlinewere designated as L13, L14, L15, L16, L17, L18 and L19. Qualitative lipolytic activity newlinewas analysed using methyl red olive oil agar medium, while quantitative lipolytic activity newlinewas done using volumetric analysis using olive oil as substrate in which L13 and L18 newlineshowed highest lipolytic activity among all isolates. Partial identification of seven newlineselected Lactic acid bacteria at species level was done using biochemical characterization newlineand conventional carbohydrate fermentation tests. newlineAll seven selected isolates gave promising result for survival and growth in acidic newlinecondition and high bile salt concentration. Growth was measured by turbidometric newlineanalysis using spectrophotometer after 2 hour and 4 hour of incubation at 37oC newlineanaerobically. L13, L15 and L17 can survive and grow moderately while L19 showed newlineefficient growth at pH 3 and pH 4 after 2h and 4h. Among all
