Abstract
Karisalai Karpam, a polyherbal formulation was subjected to various analytical techniques. Botanical parameters revealed that dark green in color, with a pungent odor, bitter taste, and fine texture. Karisalai Karpam formulator drug of Siddha medicine was safe from As, Cd, Hg, and Pb since the concentrations of those heavy metals were less than permissible limits. Phytochemical screening of Karisalai Karpam showed the presence of alkaloids, flavonoids, glycosides and terpenoids. Karisalai Karpam extracts using different solvents chloroform, methanol, acetone and ethyl acetate were studied for partial characterization by TLC. Maximum phytochemicals were visualized in chloroform extract. Result analysis of HPTLC studies on Karisalai Karpam revealed the presence of nine prominent peaks. Karisalai Karpam exhibited a powerful scavenging activity for ABTS radical cations in a concentration dependent manner. Histopathological studies of male and female rats in Sub-acute toxicity study revealed no damage in all vital organs. Liver weights of rats treated with ethanol and paracetamol (0.5ml/100gm) were significantly increased, and the normal weights were restored when after treating with the trial drug Karisalai Karpam. A result obtained from this study provides valuable preliminary data on the toxicity profile of study drug Karisalai Karpam. The drug Karisalai Karpam appears to be safe, non-toxic, causes no apparent organ damage. Collectively the findings of the present study would suggest a long term usage of this drug has no adverse effect on study animals. The results of present pharmacological study on acute and chronic model of hepatotoxic activity in rodents clearly demonstrate that the various biochemical markers like serum AST, ALT, ALP, total, direct, indirect bilirubin levels including liver weight, and histopathological alterations produced by ethanol and paracetamol in the serum and tissue were reversed significantly by the treatment with Karisalai Karpam at the dose of 200 and 400 mg/kg.
