Abstract
Drug repurposing (repositioning), which refers to the identification
newlineof new clinical indications for existing drugs, has become an important
newlinestrategy for drug development. Drug development is a lengthy,
newlinecomplicated, and highly expensive process, associated with a high degree
newlineof uncertainty that a drug will eventually succeed. In order to overcome
newlinethese challenges, drug developers are becoming more and more active in
newlineidentifying new uses for approved drugs. Computational predictions of
newlinedrug-disease associations provide a promising way to identify effective
newlinedrugs for a particular disease.
newlineNon-coding RNAs are RNA molecules that do not code for proteins.
newlineAberrant expressions of non-coding RNAs are correlated with a broad
newlinespectrum of human diseases. MicroRNAs (MiRNAs) are short non-coding
newlineRNAs that control and coordinate various biological processes. The most
newlinerecent studies in pharmacogenomics have demonstrated that drugs can
newlinetarget miRNAs and regulate their expression levels. However, few studies
newlineattempted to discover drug-disease causal relationships through the
newlinemolecular basis of miRNA. Given the fact that the inappropriate expression
newlineof miRNAs is related to many kinds of human diseases, developing drugs
newlineto target specific miRNAs and modulate their activities would be a promising
newlineapproach to human disease treatment. It offers an innovative insight into
newlinedrug repositioning. This research work mainly focused on developing a
newlinemethodology that discovers novel drug-disease associations through the
newlinetarget miRNAs. Developing miRNA-based computational models to inferpotential drug-disease associations for drug repositioning would be an
newlineeffective complement. The proposed method could successfully discover
newlinethe causality underlying drugs and diseases.

newlineas biomarkers and treatment targets for chronic diseases. Drugs achieve
newlinetheir therapeutic functions by binding with specific target molecules.
