In recent years, organometallic cobalt and ruthenium-based compounds have newlinegathered significant attention in anticancer drug discovery due to their lower toxicity and newlinebroader spectrum of activity compared to traditional platinum-based chemotherapy agents. newlineComplexes featuring pentamethylcyclopentadienyl cobalt (III) and half-sandwich Ru(II)- newlinearene structures offer vast opportunities for structural modification, which can enhance newlinetheir antiproliferative activity against cancer cells. In this thesis, various organometallic newlinecobalt and ruthenium complexes with bioactive ligands were synthesized and newlinecharacterized. Key factors such as hydrophobicity, hydrogen bonding, and ligand planarity newlinecontribute to promoting cytotoxic effects against cancer cells. Incorporating active Schiff newlinebase and pyrazolyl ligands into these complexes significantly enhances their anticancer newlineefficacy when compared to cisplatin, a widely used chemotherapy drug. However, newlinechallenges like potential toxicity and lack of biocompatibility may hinder their progress newlinetoward clinical use. To address these limitations, drug delivery systems have been newlinedeveloped to improve the therapeutic potential of cobalt and ruthenium complexes. newlineSpecifically, to polydiacetylene-supported liposomes of pentamethylcyclopentadienyl newlinecobalt(III) complexes have shown enhanced antiproliferative activity in cancer cells while newlineminimizing their toxicity to healthy cells. Additionally, modifying the organoruthenium newlinecomplex into a Ru(II)-encapsulated apoferritin nanoconjugate has improved cell newlinepermeability and increased cytotoxicity. Ultimately, the development of such newlinenanocomposites opens new avenues for interdisciplinary research in bioorganometallic newlinechemistry and nanomedicine newline
