Anti fibrotic effect of mangosteen pericarp intralesional injection in the management of areca nut induced oral submucous fibrosis in wistar albino rats model

Abstract

Oral submucous fibrosis (OSF) is a chronic, debilitating condition that newlinesignificantly compromises the quality of life through its potential for malignant newlinetransformation and severe restriction on mouth opening. The multifactorial newlineaetiology of OSF prominently features the consumption of areca nuts as a newlineprimary risk factor. Given the urgent need for effective treatments, recent newlineresearch has spotlighted the potential of natural compounds, such as newlinemangosteen (Garcinia mangostana), to modulate the fibrotic process. newlineMethods: newlineThis study explores the anti-fibrotic effect of mangosteen extract newlineadministered via intralesional injection in a Wistar albino rat model of newlineareca nut-induced OSF. The Wistar albino rats were divided into four newlinegroups to assess the effects of different treatments, including a control newlinegroup, a group treated with areca nut extract to induce OSF, and groups newlinereceiving mangosteen extract either concurrently with areca nut extract newlineor as a post-treatment. OSMF was induced using 50 mg/ml of areca nut newlineextract, administered intralesionally daily for 12 weeks. Rats with newlineinduced OSMF were then treated with mangosteen extract administered intralesionally twice weekly for three months. The study evaluated the newlineantimicrobial, anti-inflammatory, and antioxidant properties of the newlinemangosteen pericarp extract using in vitro assays and assessed its newlineimpact on human primary oral submucous fibrosis cells through cell newlineviability, migration, and gene expression analyses. The levels of newlineTransforming Growth Factor Beta (TGF-and#946;) were analysed using ELISA, newlineand statistical analysis was performed using SPSS with a significance newlinelevel set at plt0.05. The body weight and serum iron levels are evaluated newlinefor all 4 groups during 3 phases of treatment. newlineResults: newlineMangosteen pericarp extract demonstrated significant antimicrobial, newlineanti-inflammatory, and antioxidant activities in a concentration dependent newlinemanner.
