dc.description.abstractThe fruits of Psidium guajava L. was selected for the study to extract mucilage and to be used as a natural rate controlling polymer in oral sustained release tablet for sustaining the release of model drug aceclofenac. The extracted polymer is non toxic, inert, biodegradable, cost effective and easily available with easy extraction process. The sustaining/controlling action is remarkably exhibited with 100 mg polymer concentration for 24 hrs. The sustaining action is directly proportional to polymer concentration and is dependent on the type of diluent used in formulation. The presence of polysaccharides might be responsible for the sustaining property of extracted mucilage. The optimized formulation exhibited zero order drug release and Peppas kinetics model reveal that extracted polymer can be used in sustained release formulation. The sustained release of the drug is due to polymer relaxation and erosion. When comparing the in-vitro drug release of optimized formulation with marketed sustained release tablet of aceclofenac reveal that optimized formulation can be used once day formulation for delivery of aceclofenac in conditions such as rheumatoid arthritis. In comparison to other commercially available polymers, extracted polymer can also be used with the lesser or near concentration of the other natural polymers. The pharmacokinetic parameters determined in in-vivo study also propose that the extracted mucilage posses sustained release property. RECOMMENDATION: The extracted polymer (mucilage) from the fruits of Psidium guajava L. was proved for its sustained release property. The extracted polymer has got potential merits such as non toxic, inert, biodegradable, cost effective easily available polymer and can be extracted with easy extraction process. It can be recommended that this polymer from respective plant fruit will be the plant based biopolymer as a novel excipients that can be used in sustained release formulations. newline newline newline newline
