dc.description.abstractIntroduction: Ayurveda specifically defines a class of CNS modulating drugs i.e. Medhya Rasayana drugs. There are some single drug like vidhara or combination of drug like Vrudhadarak rasayan are elaborated in grantha s. Now a days it is necessary to find out the exact action of drug i.e pharmaco-kinetics and dynamics. So, Present work shows, the influence of vidhara as a single drug and Shatavari processed Vidhara (Vrudhadarak rasayana) on the differentiation of mesenchymal stem cells in to neural lineage. Aim : To investigate the effect of Vidhara as a single drug and Shatavari processed Vidhara (vruddhadarak rasayan) on mesenchymal stem cells differentiation in neural lineage. Material and methods: Pharmaceutical study revel the identification and collection of raw drug by their morphology (External apperance). Authentification reports ware done by Agharkar Research Institute. S.O.P. was developed for Shatavari processed Vidhara (Vrudhadarak rasayan) by preparing 3 batches with 7 times bhavna of Shatavari swaras in each batch. Average mean of 3 batches were calculated. Duration of 1st to 7th bhavna varies from 5 hrs to 3hrs. (Gradually decreases). Swaras requirement also varies from 2000 ml to 850 ml (Gradually decreases). Weight gain after drying of churna is 666gm i.e. 32%. Final product was a homogenous mixture with color changes from creamish brown to dark brown and pleasant smell. Standard parameters of quality control were established with the help of analytical study, which revel assessment with different modern parameters of the raw drugs and finished product. Experimental study revel the identification of stem cells by characterization of stem cells into tri-lineage differentiation. (i.e. osteogenic, chondrogenic and neurogenic differentiation.) and FACs. MTT assay was done to evaluate the effect of toxicity of drug. Neural differentiation with Vidhara and Shatavari processed Vidhara ware done with the help of DPSCs and GSCs and it shows neural differentiation in 7 days. Gene expression of DPSCs andGSCs
