Role of Brain Derived Neurotrophic Factor in Type 2 Diabetes

Abstract

The effect of CNS associated with T2D has been a subject of keen interest from newlinefew years back in different model organisms. Certain behavioral phenotypes like stress, newlineanxiety, depression, learning, memory and BDNF gene, seem to be suitable for newlineunderstanding the same in the high-sugar diet induced hyperglycemic or T2D model of newlinezebrafish, instead of simple or chemically induced T2D model. newlineThis thesis presents a keen observations/investigations/understanding on the role of newlineCNS and specifically BDNF in T2D. Investigations have been carried out using different newlinebehavioral, statistical, molecular, in-silico and meta-analysis methods to fulfill the newlinefollowing objectives: (i) To study the in-silico analysis of SNPs in human BDNF gene newlineassociated with these CNS functions; (ii) To develop high-sugar induced Type 2 Diabetic newlinemodel of Zebrafish; (iii) To study the effect of high sugar diet induced T2D on CNS newlinefunctions like stress, anxiety, depression, learning and memory using zebrafish as a model newlineorganism; (iv) To study the change in protein expression levels of BDNF in same T2D newlinemodel of zebrafish. newlineThe organization of the thesis is structured in 7 chapters. Chapter 1 presents the newlinebackground of the work in the thesis followed by a brief account of introduction to T2D newlineand BDNF and their interplay across certain CNS functions. Chapter 2 describes the meta- newlineanalysis methods for finding the association of BDNF gene SNP G196A with T2DO. newlineChapter 3 reveals the comprehensive in-silico analysis of BDNF gene SNPs in human newlineBDNF gene associated with these CNS functions. Chapter 4 proposes the development of newlinehigh-sugar diet induced hyperglycemia or Type 2 Diabetic model in zebrafish. Chapter 5 newlineexplains the effect of high- sugar diet induced Type 2 Diabetes on stress, anxiety and newlinedepression in zebrafish. Chapter 6 explains the effect of high-sugar diet induced Type 2 newlineDiabetes on the complex processes of learning and memory in zebrafish.
