dc.description.abstractABSTRACT newlineThe present research work was done to develop andamp; evaluate transdermal Patch of best newlinepharmacological active fraction of Strychnos nux-vomica seeds for wound healing newlineactivity. Strychnos nux-vomica seeds contains various phytoconstituents including newlineterpenoids like lupeol, stigmasta, 25-triene-3-ol, uvaol, and 11-oxo-amyrin palmitate newlineand polyphenols and organic acids like Chlorogenic acid, Catechol, p-hydroxy newlinephenylacetic acid, caffeic acid, and p-hydroxybenzoic acid apart from alkaloids. Often newlineStrychnos nux-vomica is known for its alkaloidal content namely brucine and newlinestrychnine. Here an attempt is made to check the therapeutic effect of other secondary newlinemetabolites which are present in its seeds. Seeds of Strychnos nux-vomica had been newlinecollected and verified. Three fractions namely Alkaloid fraction (AF), Terpenoid newlineFraction (TF) and Polyphenol Fractions (PF) were extracted with different solvents newlineusing Soxhlet extraction methods. All fractions deliver significant wound healing newlineactivity in excision and incision wound healing models in rats. When compared to the newlinecontrol group, the rats given 200 mg/kg bw of Polyphenol fraction had a high wound newlinehealing activity as compared to other groups. So, polyphenol fraction at 200mg/kg bw newlineis used for the formulation of transdermal patch. On the basis of Preliminary newlinescreening of different polymer combinations, PVP K 30 and Eudragit RL 100 was newlineselected as it shown better overall results as compared to others. No major change or newlineappearance was observed in peaks of IR-spectra of physical-mixture indicating the newlinecompatibility within extract and polymers PVP K 30 and Eudragit RL 100. Using the newlineSolvent casting approach, matrix-style transdermal patches of Polyphenol fraction newlinewere formulated. Eight formulations (F1-F8) are developed on the basis of varying newlineratios of polymers and plasticizer. Based on the appearance results, it was noticed that newlineall eight batches were found to be transparent and non-sticky in nature.
