Abstract
Rumex hastatus is considered an important traditional folk medicine and commonly used in Himachal Pradesh, India for treating many diseases. In this study, the methanol extract of leaves was evaluated for their protective effect against hepatic and cognition decline in animal models liver cirrhosis; ethanol induced hepatotoxicity and thioacetamide induced hepatotoxicity in rats.
newlineLeaves of Rumex hastatus were successively extracted using petroleum ether (PERH), chloroform (CERH), ethyl acetate (EAERH), methanol (MERH) and water (AERH). All the successive solvent extracts were subjected to preface phytochemicals screening for the finding of a mixture of phytoconstituents present in the extracts. Total Phenolic content, total flavonoids and in vitro antioxidant activity of all leaves extracts were carried out by using DPPH assay, reducing power assay, metal chelating activity, nitric oxide assay and H2O2 activity. The maximum safe dose of methanol extract of R. hastatus (MERH) was determined by acute oral toxicity studies in Wistar rats (200-250g) at 2000 mg/kg, followed OECD guideline 423 and were observed for 14 days. The protective effect of R. hastatus were investigated at dose of 100, 200 and 400 mg/kg/day orally against ethnol induced hepatotoxicity during 21 days and thioacetamide induced hepatotoxicty during 28 days in Wistar rats (200-250g). Protective effect of MERH was evaluated comparing with silymarin standard at recommended dose (25mg/kg/day) orally against ethnol induced hepatotoxicity during 21 days and thioacetamide induced hepatotoxicty during 28 days. The protective effect was evaluated using the following parameters; novel object recognition task for cognition, serum liver biochemical markers, serum ammonia estimation,liver histopathology, as well as endogenous antioxidant markers. Furthermore inflammatory markers like IL-1beta, IL-6, TNF- alpha, GABA, Glutamate and Acetylcholinestrase activity in brain tissue.
