Abstract
The present work deals with the formulation and optimization of pectin-based colon
newlinespecific microparticles and pellets containing Resveratrol. Pectin based microparticles
newlineand pellets coated with Cellulose acetate phthalate containing Resveratrol were
newlineformulated with in five different batches (MF1-MF5 and PF1-PF5). Box-Behnken design
newline(BBD) matrix was applied to optimise the core formulations. Systematic optimization of
newlineuncoated microsphere was done by Box-Behnken design (BBD) employing three critical
newlinefactors, pectin concentration (A); Stirring speed (B) and Span 80 concentration (C).
newlineParticle size (and#956;m), entrapment efficiency (EE %) and recovery (%) were the key CQAs.
newlinePectin microparticles and pellets were coated by CAP using the oil-in-oil solvent
newlineevaporation method. Characterisation of the formulations were done by determination of
newlinepercentage yield, micromeritics properties, drug loading and entrapment efficiency
newlinefollowed by particle size analysis morphological analysis using transmission electron
newlinemicroscopy (TEM) scanning electron microscopy (SEM). Drug-excipients compatibility
newlinestudy was also performed by FTIR spectrometric analysis. The formulations were
newlineevaluated by in vitro dissolution test in simulated gastric, intestinal and colonic fluid with
newlineand without 4% w/v rat caecal matter. The formulations were evaluated by in vitro
