dc.description.abstractThe most frequently occurring disease among women is breast cancer. The main motive of this present research is to optimize and synthesize eco-friendly and low-cost silver nanoparticles (AgNPs) in bio-nanocomposites (BNCs) and silver nanoparticles loaded microemulsion (AgNPs loaded ME) for oral use and evaluate its antibacterial and anticancer efficacy in cancer cell line and animal model. Gallic acid is a biological constituent found in natural fruits having strong reducing agent and a significant anticancer activity against breast cancer. A neural network (NN) optimization technique was used for synthesis of AgNPs in BNCs and determine the relationship between the formulation variables and response variables (size of AgNPs). The optimized AgNPs in BNCs formulation was characterized and evaluated for particle size and shape, morphological characeterization, particles charge and in vitro drug release studies. The pseudo-ternary phase diagram and 3-factor, 4 response D-optimal mixture model were applied for synthesis and evaluating AgNPs loaded ME. The silver nanoparticles loaded microemulsion was evaluate for physicochemical properties, particle size, shape, surace morphology, zeta potential and in vitro release studies. AgNPs in BNCs and AgNPs loaded ME exhibit the active drug release in the cancer cells which increases bioavailability and therapeutic concentration of drug to cancer cells compared to healthy cells. The synthezised AgNPs in bio-nanocomposites and AgNPs loaded microemulsion is a potential candidate for inhibiting the growth of bacteria and significant antitumor potential against MCF-7 cancer cells. The in-vivo study determines that taking AgNPs in BNCs and AgNPs loaded ME formulations caused a remarkable potential inhibition in the proliferation rate of Ehrlich ascites solid tumor and tumor mass of the mice in contrast to the tumor control. The results confirmed that AgNPs in BNCs and AgNPs loaded ME appears promising anticancer therapy regarding stability, low cost and easy preparation.
