Synthetic investigation in azetidin 2 ones and related biologically active heterocycles

Abstract

Considering the current need and biological significance of and#946;-lactams and the effect of various ring substitutions/heterocycle tethering on the pharmacological action of the molecule, here we are describing new synthetic routes for the development of newer and#946;-lactam analogues linked with different organic appendages and heterocyclic functionalities. Here we have described the development of 3-(4 -substitutedphenylthio)-and#946;-lactams which were further subjected to C-3 functionalization for nucleophilic substitution studies. In addition, a novel synthetic methodology has been described for the synthesis of stereoselective synthesis of (R)-3-phenylsulfinyl-and#946;-lactams and (S)-3-allyl-3-phenylsufinyl-and#946;-lactams. In continuation to this, novel one pot synthetic methodology for the development of (Z)- and (E)-3-allylidene-and#946;-lactams has also been described. Simultaneously, the C-3 isoxazole/dihydroisoxazole linked and#946;-lactam hybrids and 1,2,3-triazole tethered and#946;-lactam derivatives have been synthesized via regiospecific [3+2] cycloaddition methodology. Here is the description of the synthetic strategy for the formation of C-3 halohydrin-linked and#946;-lactam analogues. Besides, the development of C-3 and#945;-haloallyl linked and#946;-lactam derivatives has also been elaborated. newline
