dc.description.abstractBackground: Premna tomentosa is a medicinal plant of the Verbenaceae newlinefamily. It has been used in India for the treatment of liver disorders, diarrhoea, newlineconjunctivitis, headache, epilepsy, fever, stomachache and skin diseases. The present newlinestudy aimed to assess the hepatoprotective, Lipid-lowering and antioxidant effect of newlinethe leaf extract of Premna tomentosa. newlineMaterials and Methods: Hepatoprotective activity of Premna tomentosa was newlinestudied against 40% ethanol (1 ml/100 g b.wt p.o) induced hepatotoxicity. 36 Male newlinealbino wistar rats weighing 150-200 g divided into six groups were used for this study. newlineRats were caged under standard temperature of 24 ± 2o newlinec with a 12 h light/dark cycle. newlineThey were fed with standard Nutrient laboratory animal feed and water ad newlinelibitum.Intoxicated rats were treated with different doses of leaf extract of Premna newlinetomentosa (500 and 750 mg /kg b.wt) to select the most effective dose. The effects of newline45 days treatment with this dose (500 and 750 mg /kg) on Bodyweight, Lipid profile, newlineLiver markers (ALP, SGOT, SGPT), Renal profile (Total Urea, Blood urea nitrogen, newlineTotal Protein, Albumin, Globulin, Creatinine) and liver tissues antioxidants liver tissue newline(i.e., LPO, CAT, GPX, SOD), Histological analysis of Liver and Kidney, newlineHistochemical analysisof Liver were evaluated. newlineResults: The most effective dose of leaf extract of Premna tomentosa was EPT newline(750 mg/kg b.wt) after 45 days treatment with this resulted in significant decrease in newlineLiver markers (p lt 0.05). Hepatotoxicity produced in Lipid profile, liver markers newline(SGOT, SGPT, ALP), Renal profile (Total Urea, Blood urea nitrogen, Total Protein, newlineAlbumin, Globulin, Creatinine) and liver tissues antioxidants (i.e., LPO, CAT, GPX, newlineSOD) which were restored near to normal levels (p lt 0.05). These results were further newlinesupported by histopathological studies. newlineConclusion: Alcohol-intoxicated rats responded favorably to treatment with newlineleaf extract of Prema tomentosa which exhibits Hepatoprotective, Lipid-lowering and newlineAnti-oxidant effects
