Abstract
Psychrotrophs are cold adapted mesophilic organisms They
newlinesucceed in maintaining adequate metabolic fluxes by the production of coldactive
newlineenzymes which can efficiently catalyze biochemical reactions at low
newlinetemperature with a higher specific activity Lipases EC 3 1 1 3 catalyze the
newlinehydrolysis of triglycerides to free fatty acids and glycerol Biotechnologically
newlinecold active lipases have gained considerable importance as biocatalyst as a
newlineresult of their increasing use in the organic synthesis of chiral intermediates
newlinePsychrotropic opportunistic pathogen Serratia marcescens secretes
newlinea cold active lipase LipA that is extensively used in the production of chiral
newlineprecursor for the coronary vasodilator diltiazem In the present study attempts
newlinehave been made to understand the molecular adaptation of LipA to cold
newlineconditions Further to prove the bi functional nature of LipA displayed by
newlineamino acid sequences and finally to functionally characterize the amino and
newlinecarboxy domains of LipA
newlineLipA is the first enzyme of its family to have been studied for cold
newlineAdaptation First objective of the work was to understand the kinetic
newlineoptimization of LipA LipA was cloned expressed and purified
