dc.description.abstractBackground: Asthma is a serious health problem which has globally affected millions of newlinepeople. Lot of study has been done to examine the biological properties of Mangifera newlineindica and Nicotiana tabacum but there are no scientific reports about the outcome of the newlinetwo plants on the contractility of ASM. Objectives: This study was initiated with an aim newlineto assess the acute toxicity and the ability of hydroalcoholic extract of the two plants to newlineinhibit the spasmogen induced contraction in isolated lung strips of guinea pigs. The newlinepromising extract was fractionated using solvents of varying polarity and further newlineevaluated for their leukotriene inhibitory potential. The fraction exhibiting best activity newlinewas subjected to column chromatography and a compound was isolated and analysed newlineusing physical and spectral techniques. Methodology: The plant material was newlinestandardized and extracted using aqueous ethanol (1:1) by cold percolation method. The newlineextracts were analysed qualitatively and quantitatively for the presence of different kinds newlineof phytochemical constituents using standard chemical and spectrophotometric methods. newlineThe oral acute toxicity studies were done according to OECD guideline no. 420. A limit newlinedose of extracts at 2000 mg/kg BW was administered and animals were observed for newlinevarious wellness parameters, differences in body weight, consumption of water and food newlineas well as mortality. The leukotriene inhibitory property (relaxant effect) of the extracts at newlinesix cumulative concentrations was evaluated in isolated lung tissues of guinea pigs newlineprecontracted with LTD4. Standard drug used was montelukast and negative control newlineused was normal saline. Most promising extract was fractionated into four parts using nhexane, newlineethyl acetate, n-butanol and water as solvents. The fractions were qualitatively newlineanalysed for presence of various phytochemicals and evaluated for their leukotriene newlineinhibitory potential.
