Background: Streptococcus mutans produce glucans via newlineglucosyltransferase enzymes, aiding pathogen adhesion to teeth. It is newlineessential to investigate therapeutic agents capable of inhibiting these newlineenzymes without negatively impacting beneficial microorganisms. newlineAim: The study aimed to evaluate the anticariogenic activity of a newlinecommercially available toothpaste containing Salvadora persica newline(Miswak) compared with a commercially available fluoridated toothpaste newlineusing a randomized controlled trial. newlineMaterials and Methods: A triple-blinded, randomized controlled trial newlineincluding 88 adult participants were randomly assigned to Group A newline(miswak-containing toothpaste) or Group B (fluoride toothpaste) based newlineon a computer-generated sequence. Clinical examination, followed by newlinecollection of dental plaque was done for assessment of caries experience newlineand analysis of glucosyltransferase enzyme levels and total bacterial newlinecount. The participants were followed up after 3 and 6 months of newlinetoothpaste use. Statistical analysis evaluated the significant differences newlinebetween and within the groups. newlineResults: Both groups showed a significant decrease in newlineglucosyltransferase levels, DMFS scores, and CFU counts after 6 months newline(p=0.000). No significant difference was observed between the groups in newlineassessed outcomes.
