The role of oxidative stress sirtuin 1 level and gene polymorphism in diabetic nephropathy

Abstract

Traditionally biomarkers are used to monitor and investigate the newlineprogression of diabetic nephropathy. Recent studies showed advanced newlinebiomarkers which are significantly increased in type 2 diabetes mellitus and newlineserve as early biomarkers for glomerular and tubular functions. Apart from newlinethese biomarkers, recently genome-wide associations are discovered and newlinealso several novel genetic variants associated with diabetic nephropathy. newlineElucidating of such genes may be useful for early identification of at-risk newlineindividuals. The present study emphasizes molecular mechanism of sirtuin 1 newlinesnp s rs 3818292 and rs10823108 in diabetic nephropathy. To fill this gap, the newlinestudy was undertaken to address the impact of genetic events in diabetic newlinenephropathy in south Indian population. The study showed that SNP s within Sirtuin 1 gene are associated with susceptibility to diabetic nephropathy in indian case-control studies. Results showed that selected SNP s in SIRT 1 gene are not found to be significantly newlineassociated with diabetic nephropathy as compared to the control subjects and newlineno significant difference was observed between the genotype and allelic newlinefrequencies of SIRT1 SNPs between diabetes and diabetes nephropathy. newlineThese findings could not be very conclusive due to less number. Moreover, newlineSirtuin 1 level was decreased in diabetic nephropathy as compared to newlinediabetes revealing definite significant involvement of SIRT1 with disease newlinepathogenesis. While, MDA level was increased and SOD, GR, and GPx levels newlinewere decreased in diabetic nephropathy. newline
