Synthesis DNA Interaction and Biological Activities of Some Metal Complex Ligands with Schiff Base Derived from Substituted Salicyaldehyde and Amino Acid

dc.description.abstractTuberculosis is the ninth cause of global death, more than any other infectious disease. With growing drug resistance the epidemic remains and will require significant attention for the elimination of this disease to occur. With susceptible TB treatment not changing over the past decades and the advent of drug resistance, new drugs and regimens are required to treat the diseases. In this research, an attempt, was made to synthesize some new schiff base metal complexes with a mono-nuclear metal center, and investigated their activities against anti-microbial and anti-tubercular. Docking simulation using Auto Dock was performed to position these complexes into the active site of Enoyl CoA reductase to determine the probable binding conformation. In the present investigation, molecular docking was carried out for thirty one metal complex derivatives, out of which twenty five metal complexes with high docking score were synthesized in appreciable yield (96-70%). Molecular docking revealed the importance of electron-deficient oxygen in promoting biological activity The spectral data (FT-IR, Mass spectra, 1H NMR, 13C NMR, UV-spectra, Elemental analysis, Conductomety and Magnetic properties) of the synthesized metal complex were found in good agreement with assigned molecular structure. Results of anti-microbial studies by micro titre plate and nutrients broth methods showed that thirteen metal complexes AL2 Co, AL3 Fe, AL5 Mn, SE3 Zn, SE4 Fe, SE5 Mn, GLU2 Co, GLU3 Zn, GLU4 Fe, TYR3 Zn, TYR 4 Fe, HIS3 Zn, and HIS4 Fe were the most active in comparison to the standard drugs Ciprofloxacin and Clotrimazole. The metal complexes AL2 Co, AL3 Zn, SE2 Co, SE3Zn, GLU3 Zn, GLU3 Co, SE2 Co, GL3 Zn, TYR 2 Co, TYR3 Zn, TYR5 Mn, HIS2 Co and HIS3 Zn of eleven compounds were found to be most effective anti-tubercular agents, when studied on LJ medium and RT-PCR method compared to the standard Isoniazid. Based on the above findings, zinc, cobalt and manganese complex derivatives were found to be most effectual anti-microbial and anti-tubercular agents.
