Abstract

Neuroinflammation is a complex process involving the activation of immune cells in the brain newlineand the spinal cord in response to injury, infection, or disease. While inflammation is natural newlineand a necessary part of the immune response, chronic or uncontrolled neuroinflammation can newlinecontribute to the progression of neurodegenerative diseases. Activated glia cells, microglia and newlineastrocytes, the primary immune cells of the brain release pro-inflammatory mediators that can newlinedamage neurons and disrupt normal brain function. Neurodegenerative diseases such as newlineAlzheimer s disease, Parkinson s disease, Huntington s disease, and amyotrophic lateral newlinesclerosis are characterized by the progressive loss of neurons leading to cognitive disorders. newlineThe relationship between neuroinflammation and neurodegeneration is complex and newlinebidirectional. Strategies to modulate neuroinflammation include anti-inflammatory agents, newlineimmunomodulatory agents, and lifestyle interventions like exercise and diet, which can slow down or halt the progression of these devastating diseases. HMGB1 contributes to neuroinflammation through multiple mechanisms, including its release from activated glial newlinecells, interaction with various receptors, and induction of pro-inflammatory cytokine newlineproduction. HMGB1 inhibitors have emerged as promising candidates in the field of medicinal newlinechemistry as a potential target for various therapeutic applications such as in treating sepsis, arthritis, cancer, inflammatory diseases, etc. They offer the possibility of more targeted therapies reducing side effects associated with broad-spectrum treatments or in combination with existing therapies to enhance efficacy and reduce drug resistance newline
