In the present study, the polyherbal combinations containing selected medicinal newlineplants were collected from different geographical areas and authenticated from newlineNISCAIR, New Delhi, India with different authentication nos. As per the phytochemical newlineinvestigation alkaloids, carbohydrates, flavonoids, tannins and phenolic compounds newlinewere found to be present in the various extracts of plant part and polyherbal newlinecombinations. The total phenolic content was found to be 34.4±0.10 and 27.6±1.20 mg newlineGallic acid equivalent (GAE)/g, total flavonoid contents, total tannin contents were newline24.7±0.25 and 18.1±1.20 QE/g 12.31±0.25 and 9.48±1.85 TAE/g respectively of polyherbal newlinecombinations (HAF-I and HAF-II). On the basis of TLC studies at solvent system toluene: newlineethyl acetate: formic acid (4:5:1) in TLC mainly showed Rf values were 0.46, 0.53, 0.65 newlineand 0.75 and for HPTLC analysis solvent system were choosen as cyclohexane: ethyl newlineacetate: formic acid (4:5:1) showed Rf value for HAF-I were 4.2, 5.3, 6.8, 7.6 and for newlineHAF-II were found 4.0, 4.2, 6.2, 6.5 and 6.7 revealed reported phytochemicals like newlinegallic acid, catechin, catechol and other polyphenolic presented and these presence newlineconfirmed its anti-oxidant, hepatoprotective and nephroprotective actions. The In-vitro newlinefree radical DPPH activities found to 74.17±0.18 and 75.30±0.18, Nitric Oxide newlineantioxidant activity were found to 75.3±1.10 and 76.17±1.24 at maximum with compare newlineto ascorbic acid and rutin as a reference at concentration of 100 µg/ml. The anti-oxidant newlineactivities were gradual increased with respect to increase the concentrations of different newlineparts of plant extract of medicinal plants. newlineAcute toxicity study was performed according to OECD guidelines No. 423. newlineSprague Dawley female rats were grouped into three (10 ml/kg, 2000 mg/kg and 2000 newlinemg/kg body weight) containing five animals in each. The sub-acute toxicity studies of newlinethe combinations were determined as per OECD guideline 407. Selected animals were newlinegrouped for daily oral administration of HAF-I and HAF-II at an individual dose of 200 newlineand
