Abstract
The investigation entitled Morphological, biochemical and molecular characterization of
newlinemango (Mangifera indica L.) germplasm was performed at HRC, Patharchatta and CBSH, G. B.
newlinePant University of Agriculture and Technology Pantnagar and MBGE Laboratory of UCB, Haldi,
newlinePantnagar, U.S. Nagar, Uttarakhand during the year 2021 and 2022. The whole study was completed
newlinein two different experiments. In the 1st experiment the thirty-mango germplasm maintained at HRC,
newlinePatharchatta were estimated for their morphological and biochemical characters. In the 2nd
newlineexperiment the same mango germplasm were characterized at molecular level using SSR s marker.
newlineLate fruit maturity (2nd fortnight of August) and maximum fruiting duration (121.50 days)
newlinewere observed in germplasm PMSS-1. Whereas, the early maturity (2nd-3rd week of June) was
newlinereported in PMGC-163. The maximum fruit length (15.16 cm), diameter (9.01 cm) and weight
newline(422.21 g) were observed in PMSS-31, PMGC-20 and PMGC-5, respectively. PMSS-18 (0.62 kg m-
newline3) has come out as most productive germplasm due to its maximum yield efficiency and it was
newlinefollowed by Dashehari (0.60 kg m-3) and PMSS-1 (0.60 kg m-3). In terms of fruit quality, PMGC-48
newlineexcelled in terms of TSS content (22.45 °Brix), total sugars (15.94 %) and non-reducing sugars
newline(11.65 %), while, PMGC-96 witnessed minimum titratable acidity (0.15 %). Langra was reported
newlinesuperior with regard to ascorbic acid (63.22 mg/100g), total carotenoids (7.08 mg/100g) and
newlineflavonoids (42.59 mg QE/100g) among all other mango germplasm. Germplasm PMSS-11 had
newlineperformed better in terms of total phenols (94.39 mg GAE/100g) and antioxidant activity (257.70
newlinemg TE/100g).
newlineIn the molecular study, all 25 SSR primers amplified 83 alleles and all bands (83) were
newlinepolymorphic. The SSR marker MiIIHR15a and QGMi009 produced maximum number of 5 alleles
newlinefollowed by MiIIHR17a, LMMA-8, QGMi011, MGDSSR-22, MGDSSR-25, MngSSR-14,
newlineMngSSR-24 and SSR-16 which produced 4 alleles, respectively. The amplified fragments ranged
newlinefrom 125-400 bp. The polymorphic information content (PIC) was calculated for each primer and it
newlinewas ranged from 0.358-0.746. The highest PIC value was noted from MiIIHR15a (0.746) and the
newlinelowest for MGDSSR-11 (0.358). Germplasm identified as promising in the investigation may prove
newlineto be an important genetic resource for mango improvement programmes. Hybrid breeding
newlineprogrammes can use SSR diversity as a criterion to select parents when crop improvement of
newlinemango.
