Investigations on Bioactivity of Some Indian Plants for the Management of Rheumatoid Arthritis

Abstract

The study was designed to investigate the anti arthritic potential of three selected plants, a formulation from those bioactive extracts and isolation of molecule from bioactive fractions. newlineAll the selected crude drugs were authenticated, pulverized and processed. The extracts were prepared by using successive solvent extraction method with increasing polarity of solvents. The obtained extracts were screened by preliminary phytochemical tests to categorically chemoprofile them. Then the screened extracts e.g. EAF, WFP, WFE PHC and PHE we reevaluated for antiarthritic action by complete Freunds adjuvant CFA induced arthritis in adult Lewis rats 150 200gm after acute toxicity study. Anti arthritic activity of EAF 10 mg kg, WFP 100 mg kg, WFE 100mg kg, PHC 100mgkg and PHE 100mgkg all orally was evaluated in CFA induced arthritic rats for prophylactic and therapeutic effect. Paw volume, body weight changes, locomotor activity, hemoglobin Hb count, erythrocyte sedimentation rate, LPO, and SOD levels were assessed in both developing and developed phases of arthritis. These were supported by histological and radiological studies of the affected joints. Statistical analysis was performed using oneway ANOVA followed by Dunnett s test. P 0.05 was considered statistically significant A suspension with bioactive extracts was formulated and evaluated for antiarthritic action as mentioned above. The compounds were isolated from active fraction by solvent-solvent extraction technique and were characterized by spectroscopic and chromatographic techniques. newline newlineThe EAF 10mg kg, WFP 100mgkg and PHE 100 mg kg p.o. showed significant reduction in paw volume, body weight, ESR, and LPO level. There was a significant increase in hemoglobin count, locomotors activity, and SOD level. The suspension formulation F2 FAWP prepared from EAF, WFP and PHE showed acceptable and agreeable physicochemical characteristics than otherformulations. newline newline newline
