Abstract
Background:
newlineOral sub mucous fibrosis (OSMF) is a condition wherein mesenchymal response to injury
newlineare mediated through changes in fibroblast structure and function. Fibroblasts differentiate
newlineto alpha smooth muscle actin positive (and#945;SMA+) myofibroblast, which is key feature
newlineunderlying pathogenesis of OSMF. Studies highlight that myofibroblast phenotypes
newlinevary from physiological to pathological conditions. Recently, role of transforming
newlinegrowth factor and#946; (TGFand#946;) and mitochondrial reprogramming has been studied in organ fibrosis.
newlineIn OSMF, the role of arecoline and TGFand#946; is well-recognized but there is a paucity
newlinein literature regarding their effect in myofibroblast differentiation and mitochondrial
newlinefunction. Thus, the present work aimed to evaluate the role of arecoline and TGFand#946; in
newlinemyofibroblast differentiation by study of mitochondrial bioenergetics.
newlineObjectives:
newlinePrimary Objective: To evaluate role of arecoline and TGFand#946; on myofibroblast transdifferentiation
newlineusing human oral fibroblast cell lines.
newlineSecondary Objectives:
newline1. To assess the effect of arecoline, TGFand#946; and combination of arecoline and TGFand#946; on
newlinemyofibroblast transdifferentiation.
newline2. To assess the mitochondrial bioenergetics in myofibroblast transdifferentiation
newlineinduced by arecoline, TGFand#946; and combination of arecoline and TGF and#946;.
newline3. To compare the mitochondrial bioenergetics in myofibroblast transdifferentiation
newlineinduced by arecoline, TGFand#946; and combination of arecoline and TGFand#946;
