Enumeration of the pathophysiological basis of antiinflammatory and antiangiogenic cellular responses in milieu with bio functional properties of Memecylon malabaricum kostel

Abstract

To evaluate the anti-inflammatory properties of a novel compound Memecylaene, isolated fromMemecylon malabaricum(Cl.) in acute and chronic inflammatory models. Memecylaene was tested for antiinflammatoryactivity in albino rats. The in vivo anti-inflammatory activity was studied using acute and subacuteanimal models. The biochemical parameters such as antioxidant enzyme activities from ganuloma, lipidperoxidation inhibition in the liver of granuloma induced rats; as well as mucopolysaccharides from thegranuloma were carried out. The in vitro effect of memecyleane was evaluated through the inhibition of snakevenom PLA 2s which are known to induce inflammation. Memecylaene exhibited significant anti-inflammatoryactivity in acute and sub-acute models of inflammation with significant (Plt0.001) reduction of paw edema andgranuloma tissue compared to treatment with standard drug diclofenac sodium. Acute oral toxicity studyindicates that the molecule is safe as drug. Memecylaene treatment significantly increased the antioxidantenzyme activities (CAT, SOD and GPx (Plt0.05). Inhibition of lipid peroxides in liver and mucopolysaccharides ingranuloma tissue was found. Memecylaene inhibited the PLA2 s V and VIII of Russell viper venom with an IC50value of 12.58and#956;g and10and#956;g. Memecylaene showed anti-inflammatory activity in both the models ofinflammation which is attributed to their antioxidant and phospholipase A2 inhibitory activities. Thus, thestudy validated the scientific rationale of ethno medicinal use of M. malabaricum to inflammatory associateddiseases and unveils its mechanism of action.
