Abstract
Current research is designed to develop the DNA sensor-based point-of-care device for the
newlinedetection of scrub typhus. Scrub typhus is a deadly disease transmitted by mites and caused by
newlinethe bacteria Orientia tsutsugamushi (O. tsutsugamushi). It is a reappearing disease of the
newlinetsutsugamushi triangle transmitted by the bite of chiggers (larval stage). Scrub typhus is difficult
newlineto diagnose as its symptoms are similar to other fever-related illnesses. Enzyme-linked
newlineimmunosorbent assay (ELISA), Immunofluorescence assay (IFA), Immunochromatographic test
newline(ICT), Weil-Felix, Polymerase chain reaction (PCR), and Loop-mediated isothermal
newlineamplification (LAMP) are all effective diagnostic procedures for scrub typhus. The Weil-Felix
newlinetest was first employed in poor nations to screen for scrub typhus, but it was not recommended
newlinedue to its lack of specificity and sensitivity. Other antibody-based methods such as IFA and
newlineELISA are mostly used in the diagnosis of scrub typhus due to their high sensitivity and
newlinespecificity, but they do not have the ability to detect infection in the early stages and require
newlineconvalescent sampling for confirmation. On another deed, PCR-based methods combating the
newlinelimitation of current diagnosis methods and are able to detect the infection at early stage with
